Confocal Microscope Applications:

Several images generated by means of shifting the focal plane can be combined into a 3D image stack, and be projected to a stereo image.

Photo-manipulation experiments can be performed by confocal microscope. By the aid of the Acousto Optical Turnable Filter (AOTF) and the scanners, an imaging region and a stimulating region can be defined. Through a photo-stimulation program set by the software, a time-lapse image stack will be generated, so that the intensity change before and after photo-stimulation can be analyzed. These kind of experiments includes: FRAP (Fluorescence recovery after photo-bleaching), FLIP (Fluorescence loss in photo-bleaching), iFRAP (inverse FRAP), FLAP (Fluorescence localization after photo-bleaching), Photo-activation, Reversible photo-activation, etc.

Several off-line or on-line analysis can be performed by the confocal software, it includes: area, threshold area, intensity, histogram, profile, colocalization, FRAP, FRET, etc.
